| Background | Methionine adenosyltransferase II alpha (MAT2A) uses methionine and ATP as substrates to generate S-adenosylmethionine (SAM), a universal methyl group donor in cells. Tumor-initiating cells (TICs) have high methionine cycle activity mediated by MAT2A, leading to their metabolic dependency on exogenous methionine (1). Studies show that mTORC1 signaling induces MAT2A expression, leading to elevated synthesis of SAM. In addition, mTORC1 increases the protein level of Wilms tumor 1-associating protein (WTAP), a positive regulatory subunit of the human N6-methyladenosine (m6A) RNA methyltransferase complex. mTORC1, therefore, promotes N6-methylation of mRNA to stimulate protein synthesis and cell growth (2). Furthermore, inhibition of MAT2A activity was shown to decrease proliferation of cancer cells deficient in methylthioadenosine phosphorylase (MTAP) (3). |
