| Background | Synaptobrevin-2 homolog Ykt6 is a member of the soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) protein family. SNARE proteins represent the core membrane fusion machinery that drives membrane fusion in the cell. Ykt6 is an unusual SNARE as it lacks a transmembrane domain but uses farnesylation and palmitoylation as membrane anchors to mediate SNARE-dependent membrane fusion (1). Ykt6 plays a key role in ER-Golgi trafficking but can also facilitate other organelle transport, including intra-Golgi, endosome-Golgi, and autophagosome formation trafficking. Ykt6 is also involved in trafficking of proteins to lysosomal compartments from the Golgi under normal conditions and in response to cellular stress. α-Synuclein, a protein that accumulates and is genetically linked to Parkinson’s disease and dementia with Lewy bodies, associates with and deactivates Ykt6 (2). Deactivation of Ykt6 by α-Synuclein alters normal α-Synuclein proteostasis and may represent a cellular mechanism by which α-Synuclein accumulation occurs. |
