Histones are highly conserved proteins that serve as the structural scaffold for the organization of nuclear DNA into chromatin. Histone modifications regulate DNA transcription, repair, recombination, and replication. Histone H2B is one of the core components of the nucleosome, the smallest subunit of chromatin. DNA base pairs are wrapped around an octamer of core histone proteins consisting of two each of Histone H2A, H2B, H3 and H34. The most commonly studied modifications are acetylation, phosphorylation, methylation, and ubiquitination. A number of studies have implicated ubiquitinated H2B in heterochromatic gene silencing and in the control of gene expression. Histone H2B is ubiquitylated at Lys120 by RAD6 E2 protein in conjunction with RNF20/RNF40 and this ubiquitinylated form is found preferentially associated with the transcribed regions of highly expressed genes. |
