| Background Information | The Rab family of proteins are a member of the Ras superfamily of small G-Proteins. At least 60 isoforms have been identified. These GTPases are primarily involved in regulating distinct steps in vesicle trafficking by regulating the dynamic assembly and disassembly of the multi-protein scaffolds that are involved in vesicle trafficking. In its active, GTP-bound state, the Rab protein recruits effector proteins to the membrane. Rabs lack intrinsic GTPase hydrolysis activity and thus rely on guanine nucleotide exchange factors (GEFs) and GTPase activating proteins (GAPs) to regulate their activity. Through these effectors, they regulate vessicle formation and movement. Rab4 and Rab11 are involved in internalization, sorting and transport to lysosomes or recycling of several membrane proteins, including GPCRs, GLUT-4, Aquaporin-2 or the H+/K+ ATPase. |
