| Background Information | Tribbles homolog 1 (UniProt: Q96RU8; also known as TRB-1, G-protein-coupled receptor-induced gene 2 protein, GIG-2, SKIP1) is encoded by the TRIB1 (also known as C8FW, GIG2, TRB1) gene (Gene ID: 10221) in human. TRB-1 is an evolutionary conserved monomeric protein that acts as a pseudokinase. It is widely distributed, but highest concentrations are found in the bone marrow and the thyroid gland. In mammals, it controls several interacting signaling pathways, including MAP kinases and PI-3 kinase pathways. In the N-terminal region it contains a PEST region and a pseudokinase domain. In the C-terminal region it has a COP1-binding region (aa 355-360). Due to the lack of a functional ATP-binding site it is catalytically inactive but serves as an adapter or scaffold that facilitates assembly of other proteins. Its COP1-binding motif is masked by autoinhibitory interactions with the protein kinase domain. TRB-1 has been established as an oncogene in acute myeloid leukemia and his reported to play a pivotal role in the differentiation of anti-inflammatory M2-macrophages in the pathogenesis of early atherosclerosis and during adipose tissue inflammation.
