| Background Information | Tight junctions are complexes of proteins that create intercellular boundaries between the plasma membrane domains of epithelial and endothelial cells. Many of the tight junction-associated proteins are members of the membrane associated guanylate kinase (MAGUK) family and include occludin and zona occludin family members ZO-1, ZO-2 and ZO-3. These proteins are thought to have both structural and signaling roles, and are characteristically defined by three protein-protein interaction modules: the PDZ domain, the SH3 domain and the guanylate kinase (GuK) domain. ZO-1 forms complexes with either ZO-2 or ZO-3. In addition, these proteins can also associate with claudin, occludin and F-actin, at tight junction stands, where they provide a linkage between the actin cytoskeleton and the tight junction. ZO-1 expression is significantly reduced in many breast cancer lines. ZO-2 and ZO-3 are ubiquitously expressed within epithelial tight junctions, and unlike ZO-1, which is also expressed at cell junctions of cardiac myocytes, ZO-2 is not expressed in nonepithelial tissue. |

| Entrez Gene Number |
|
| Entrez Gene Summary | This gene encodes a protein located on a cytoplasmic membrane surface of intercellular tight junctions. The encoded protein may be involved in signal transduction at cell-cell junctions. Two transcript variants encoding distinct isoforms have been identified for this gene. [provided by RefSeq]. |

| UniProt Summary | FUNCTION: The N-terminal may be involved in transducing a signal required for tight junction assembly, while the C-terminal may have specific properties of tight junctions. The alpha domain might be involved in stabilizing junctions. SUBUNIT STRUCTURE: Interacts with BVES (via the C-terminus cytoplasmic tail). Interacts with HSPA4 and KIRREL1 By similarity. Homodimer, and heterodimer with TJP2/ZO-2 and TJP3/ZO-3. Interacts with occludin, claudins, CGN/cingulin, CXADR, GJA12, GJD3 and UBN1. SUBCELLULAR LOCATION: Cell membrane; Peripheral membrane protein; Cytoplasmic side. Cell junction › tight junction. Note: Movement of ZO-1 from the cytoplasm to membrane is an early event occurring concurrently with cell-cell contact. TISSUE SPECIFICTY: The alpha-containing isoform is found in most epithelial cell junctions. The short isoform is found both in endothelial cells and the highly specialized epithelial junctions of renal glomeruli and Sertoli cells of the seminiferous tubules. DOMAIN: The second PDZ domain mediates interaction with GJA12 (By similarity). SEQUENCE SIMILARITIES: Belongs to the MAGUK family. Contains 1 guanylate kinase-like domain. Contains 3 PDZ (DHR) domains. Contains 1 SH3 domain. Contains 1 ZU5 domain. SEQUENCE CAUTION: The sequence AAA02891.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended. |
