| Background Information | Cleavage and polyadenylation specificity factor subunit 5 (UniProt: O43809; also known as Cleavage and polyadenylation specificity factor 25 kDa subunit, CFIm25, CPSF 25 kDa subunit, Nucleoside diphosphate-linked moiety X motif 21, Nudix motif 21, Pre-mRNA cleavage factor Im 25 kDa subunit) is encoded by the NUDT21 (also known as CFIM25, CPSF25, CPSF5) gene (Gene ID: 11051) in human. CFlm25 is a homodimeric protein that is a component of the cleavage factor Im (CFIm) complex, which plays a key role in pre-mRNA 3-processing. It is involved in association with CPSF6 or CPSF7 in pre-MRNA 3-end poly(A) site cleavage and poly(A) addition. The interaction of CFlm25 with the RNA is one of the earliest steps in the assembly of the 3 end processing complex and facilitates the recruitment of other processing factors. It contains one Nudix hydrolase domain (aa 76-201). The region containing aa 2-147 is considered to be essential for RNA-binding. It can be acetylated by p300/CBP and acetylation results in a reduction in its interaction with Poly(A) polymerase alpha (PAPAO). |
