| Background Information | Phosphatidylinositol 3,4,5-trisphosphate 5-phosphatase 1 (EC 3.1.3.86; UniProt Q92835; also known as hp51CN, Inositol polyphosphate-5-phosphatase of 145 kDa, p150Ship, SH2 domain-containing inositol 5-phosphatase 1, SH2 domain-containing inositol phosphatase 1, SHIP-1, SIP-145) is encoded by the INPP5D (also known as SHIP, SHIP1) gene (Gene ID 3635) in human. SHIP1 and SHIP2 are related phosphatases that play important roles in modulating cell signaling by regulating the phosphorylation state of phosphatidylinositol phosphates (PIPs). While SHIP1 and SHIP2 share a high degree of amino acid conservation, they differ significantly in their cellular expression and receptor recruitment. SHIP1 is primarily confined to cells of the hematopoietic lineage, although it is also expressed by embryonic and mesenchymal stem cells (ESCs and MSCs). SHIP2 is ubiquitously expressed in all cell and tissue types, with especially high levels found in the heart, skeletal muscle, and placenta. SHIP1 functions as s negative regulator in immunoreceptor signaling, in hematopoietic progenitor cell proliferation/survival, and as an inducer of cellular apoptosis. Humna SHIP1 consists of an N-terminal SH2 domain (a.a. 5-101) that mediates interactions with a large number of intracellular signaling proteins. SHIP1 also contains three SH3-interacting regions (a.a. 124-129, 969-974, 1040-1051) characterized by the presence of the PxxP motifs and two NPXY motifs (a.a. 912-915 & 1019-1022) that interact with SH2 domain-containing proteins. |
