| Background Information | Homeobox protein Hox-C10 (UniProt Q9NYD6; also known as Homeobox protein Hox-3I) is encoded by the HOXC10 (also known as HOX3I) gene (Gene ID 3226) in human. HOXC10 belongs to a family of highly conserved homeobox transcription factors that play an important role in morphogenesis in all multicellular organisms. Mammals possess four similar homeobox gene clusters, HOXA, HOXB, HOXC and HOXD, with HOXC genes located in a cluster on chromosome 12. Hox-C10 is also implicated to be involved in replication, where it binds in a structure-dependent manner to origins of replication and contributes to the assembly of replicative complexes. HOXC10 transcription is reported to be silenced in two estrogen deprivation-resistent cell lines, C4-12 and LTED, derived from MCF-7 breast cancer cells as a result of epigenetic hypermethylation as well as additional repressive histone modifications. Evidence indicates that Hox-C10 expression is repressed by estrogen in vitro and in vivo to promote tumor growth. Short-term estrogen withdrawal restores Hox-OXC10 expression to induce growth arrest and apoptosis, but its long-term absence results in permanent repression as a result of epigenetic methylation and the development of estrogen-independent cancer cells. |
