| Background Information | Ras-related protein Rap-1A (UniProt: P62834; also known as EC:3.6.5.2, C21KG, G-22K, GTP-binding protein smg p21A Ras-related protein Krev-1, Rap1A) is encoded by the RAP1A (also known as KREV1) gene (Gene ID: 5906) in human. The Ras superfamily is a large group of proteins that cycle between GDP- and GTP-bound forms and interact with proteins depending on the nucleotide that is bound. Two isoforms of Rap1 have been described that are known as Rap1A and Rap1B. They differ in only a few amino acids. Rap1 binds either GDP or GTP, and the change between the two states represents a molecular switch where the GTP-bound form is on and the GDP-bound form off. Rap1A, a cell membrane, lipid anchor protein, is a ubiquitously expressed protein that was first identified as a ras-tumor-suppressor gene in ras-transformed NIH 3T3 fibroblasts. It is synthesized with a short propeptide (aa 182-184) that is subsequently cleaved off to produce the mature, functional protein that has three GTP-binding sites. It is activated by guanine nucleotide-exchange factors (GEF) EPAC and EPAC2 in a cAMP-dependent manner. Rap1A counteracts the mitogenic function of Ras, at least partly because it can interact with Ras GAPs and RAF in a competitive manner.
