The Cas9 nuclease forms a stable ribonucleoprotein (RNP) complex with the guide RNA (gRNA) component. With the help of two nuclear localization signals (NLS) expressed with the Cas9 nuclease, the RNP complex enters the nucleus and cleaves target gene.
